Zusammenfassung

Ischemic stroke is the main cause of disability and an immense burden both on the resources of families and societies. Actual treatments are neuroprotective therapies only available for 3-7% of stroke patients. Consequently, new therapies are necessary to reduce the impact of ischemic stroke. In this Thesis we hypothesized that is possible to reprogram astrocytes to neurons using siRNAs for the inhibition of NOTCH1 pathway. This strategy could allow to reprogram astrocytes of the glial scar as a suitable treatment for the ischemic stroke. Therefore, we tested this treatment in an animal model of ischemic stroke. We also hypothesized that this treatment could be delivered in a tranlational way by the used of biodegradable non-toxic particles.
